CORE ANSWER

A peptide specification uses different measurements to answer different questions: what is the material, what does its purity result show and how much target material is present?

Identity, molecular mass, chromatographic purity, content and lot traceability each describe part of the record. No single number replaces the full analytical record.

01

Sequence, identity and molecular mass

The amino-acid sequence is the precise order of residues in a peptide. Molecular mass is the mass expected from that sequence and any specified chemical modifications.

Mass spectrometry can support identity by comparing measured mass-to-charge information with the mass expected for the target molecule. [1]

02

Purity and peptide content

Chromatographic purity reports the relative detector signal attributed to the target peak under the stated method. It is not automatically the mass fraction of the target material. Peptide content is a separate measurement whose method and reporting basis should be specified. [2]

A vial can therefore have a high chromatographic purity result while peptide content, total fill quantity or molecular identity remains a separate question.

03

Freeze-dried material and reconstitution

Lyophilised means that water has been removed from a frozen material under reduced pressure. Reconstitution is the controlled addition of a suitable laboratory diluent to return that dry material to solution. [3]

These terms describe physical state and handling. They do not establish identity, purity, sterility or clinical suitability.

04

Batch, lot, COA and traceability

A batch or lot number connects a physical material to its production and analytical records. A Certificate of Analysis (COA) summarises specified tests and results for a sample or lot.

  • Lot or batch identifier
  • Sample identity
  • Laboratory or testing source
  • Analytical method
  • Test date
  • Specification and reported result

Traceability depends on those elements linking coherently. A generic report with no meaningful connection to the physical lot provides weaker documentation than a lot-specific record.

KEY PRINCIPLE

Identity, purity, content and traceability are separate analytical concepts. Strong documentation makes those distinctions visible.
