How to Interpret Peptide Purity

What a reported purity percentage means, what it does not mean and why identity, content and lot traceability remain separate questions.

CORE ANSWER

A peptide purity percentage usually describes the relative proportion of the main chromatographic peak under a particular analytical method. It is not the same measurement as peptide content, molecular identity, total fill quantity or sterility.

01

What the percentage is actually describing

In a chromatographic purity test, the detector records multiple peaks. The principal peak associated with the target material is compared with other detected peaks to estimate relative purity.

The result is tied to the method, detector, integration settings and compounds that are detectable under those conditions.

02

Purity is not peptide content

Peptide content measures the amount of peptide material. Chromatographic purity reports the relative detector signal attributed to the target peak under the stated method. That signal proportion is not automatically a mass fraction. [1]

A high purity result cannot be used on its own to calculate the amount of peptide in a vial. [1]

Quantification requires a suitable calibrated assay, which may use HPLC or LC-MS. [1]

03

Identity remains a separate question

A clean chromatogram does not automatically prove that the principal peak is the intended peptide. Identity testing such as mass spectrometry can provide a separate line of evidence by comparing molecular mass with the expected compound.

04

Other quality dimensions remain outside the percentage

  • Peptide content or assay quantity
  • Molecular identity
  • Sterility
  • Endotoxin level
  • Residual solvents or other material-specific impurities
  • Stability over time

A useful purity claim should therefore sit within a wider analytical and traceability record.

KEY PRINCIPLE

Purity is one measurement. Quality interpretation improves when purity, identity, content and batch traceability are kept separate and then read together.

Evidence & Documentation

Read purity in the context of a lot record

Use Batch Records to review published analytical documentation rather than treating a headline purity percentage as the entire quality record.
